Abstract:

Traditional Flow Cytometers can result in valuable samples and time being lost due to the necessity of optimising amplifier gains and/or PMT voltage adjustments which cannot be changed post analysis.

The poster discusses how the Accuri C6 Flow cytometer utilises 24 bit high resolution digital processing technology to enable a system which operates with pre optimised detectors calibrated to operate in linear range over 7 log decades, and a software ‘zoom' function which enables exploration of all populations; resulting in a system capable of analysing a large variety of samples and ensuring that all data is captured and never lost due to incorrect set up.

Abstract:

The University of Pittsburgh Cancer Institute Cytometry Facility at the Hillman Cancer Center served as a beta site for the new Beckman Coulter Navios/Galios analytical cytometer platform.  The Navios is a 3-laser, 10-color high speed analyzer with several unique features, including both low and wide angle forward scatter parameters and novel baseline restoration electronics designed to increase sensitivity.  In this brief report we describe our experience with the Navios, including recommendations for instrument setup and panel design.  Simultaneous analysis of rare circulating endothelial cells and hematopoietic progenitor cells is presented as an example of the use of multi-color flow cytometry for the detection of rare events.  The Navios cytometer was notable for its enhanced sensitivity, wide angle forward light scatter and ease of operation.

Abstract:

Human mesenchymal stem and progenitor cells are a potentially important cellular source for cell therapy and tissue engineering. Mesenchymal progenitor cells (MPCs) are normally cultured in medium containing fetal bovine serum (FBS). However, the presence of animal proteins or serum in the expansion medium is a cause for concern when the cells are to be used in clinical applications because of the increased risk of both immune rejection of transplanted populations and disease transmission. This poster describes the development of a humanized, serum- and animal component-free culture system which promotes superior clonogenic growth and long-term expansion whilst maintaining multi-lineage differentiation potential of human bone marrow-derived mesenchymal cells.  The ability to culture human MPCs in the absence of serum and other animal-derived components will enable further development and use of MPCs populations for clinical cell therapy and tissue engineering applications.
